THE INFLUENCE OF THE COMPLEX OF PLANT POLYPHENOLS, VITAMIN K AND MINERALS IN THE STATE OF PERIODONTAL TISSUES OF RATS MODELING THE CONDITIONS OF PERIODONTITIS

Abstract

It was studied the effects of the complex of plant polyphenols of Achillea millefolium, vikasol and minerals of the Duovit during the experiments on 28 female-rats, 1,5 months age. Intact group consisted of 12 rats (I group). In the 2nd group (8 rats) was modeled periodontitis by the summary introduction of the warfarin and kuprenil. In the third group (8 rats) per os introduced the complex of the polyphenols of Achillea millefolium, vikasol and minerals in the conditions of modeling the periodontitis.

Modelling of periodontitis caused an increase of the bone resorption of the alveolar bone; tendency to reducing the content of GAG; gain of the peroxide products at insufficient functioning of antioxidant enzymes in periodontal tissues. The complex of plant polyphenols, minerals and vikasol made up the disturbed in periodontitis structural and functional state of the periodontal of rats. It had a positive anabolic effect on periodontal tissue.

The purpose of the experiment was to study the protective effects of the complex preparation of polyphenols herb Yarrow, vikasola and minerals on the condition of periodontal tissues of rats in conditions of modeling of periodontitis.

We have made a model of the total experimental periodontitis by oral administration of the antagonist of vitamin K – warfarin and kuprenil having complexing properties in respect of metal ions. In addition, kuprenil produces a multiple effect on the metabolism of collagen, its cross-linking fibers and inhibiting synthesis.

Materials and research methods. It was studied the effects of the complex of plant polyphenols of Achillea millefolium, vikasol and minerals of the Duovi during the experiments on 28 female-rats, 1,5 months age . Intact group consisted of 12 rats (I group). In the 2nd group (8 rats) was modeled periodontitis by the summary introduction of the warfarin and kuprenil. In the third group (8 rats) per os introduced the complex of the polyphenols of Achillea millefolium, vikasol and minerals in the conditions of modeling the periodontitis.

Rats were sacrificed by total bloodletting from the heart, carried out under anesthesia (sodium thiopental 40 mg/kg). Pre-separating the gums, but also identifying the upper and lower jaw, singled out the liver. Dedicated jaws were subjected to morphometric study. Objects of biochemical studies were blood serum, the supernatant of homogenates of the liver, gums and alveolar bones. The supernatant was obtained by centrifugation in the centrifuge RS-6 for 15 minutes at 3000 rpm at 4°C.

Results of researches and their discussion.

The analysis of the effect of the drug CFT with vitamin K3 and minerals of the drug Duovit were performed in simulation periodontitis total introduction of warfarin and kuprenil.

Chronic administration of warfarin and kuprenil in the duration of the 55 days led to an increase of resorption of bone structures of the periodontium.

Strengthening osteoreparative processes in the alveolar bone of the jaws preceded activation of osteoclasts, increase activity of acid phosphatase in the object of study in 3 times as a measure of reactivity of osteoclasts, the marker enzyme is acid phosphatase. The increased activity of the proinflammatory enzyme acid phosphatase in the soft tissues of the periodontium 2.5 times says about the increased inflammatory phenomena in the simulation of periodontitis.

The concentration of sialic acid was increased by 24 %: 2,22±0,24 mmol/l compared with intact group: 1,79±0,27 mmol/l increase in the level of sialic acids showed the destruction of glycoproteins CT in inflammation. The content of glycosaminoglycans – the main substance of MICRONS of connective tissue decreased as a result of simulation of periodontitis: the gum – 26 %.

Under the influence of the orally administered complex which is a polyphenol yarrow, menadione & minerals drug Duovit, bone resorption in periodontal conditions modeling of periodontitis decreased significantly: by 18% in the lower jaw and by12 % – in the upper jaw. The activity of acid phosphatase in alveolar bone under the effect of the complex decreased in 2.6 times. The content of glycosaminoglycans in periodontal tissues was significantly increased compared with the control group: in the gum level of glycosaminoglycans was increased 1.5 times more, and on the periodontis bone 2 times more.
